چکیده

The SCE2£ gene was isolated by complementing a MAla cdc2 6 ura3 strain with a S. cerevisiae wild type genomic library in the vector pFLl (1). The SCD2 6 gene maps in the vicinity of CEC2£ (chromosome 6, right arm) (2) and complements the temperaturesensitive rrir:26 mutation (group H) (3) upon integrative gene duplication. The SCD2 6 gene is not essential for viability and encodes a globular hydrophilic 17 kD protein (151 amino acids, calculated pi 5.1) containing a central cluster of acidic residues. The codon usage indicates a low expression rate, and the predicted polypeptide sequence is not related to 5415 protein sequences compiled in the PIR data base. The SCD26 gene might encode a regulatory protein related to the still unidentified CDC26 gene product.

Distinct subunit functions and cell cycle regulated phosphorylation of 20S APC/cyclosome required for anaphase in fission yeast.

We show here that the fission yeast gene products Cut9 and Nuc2 are the subunits of the 20S complex, the putative APC (anaphase promoting complex)/cyclosome which contains ubiquitin ligase activity required for cyclin and Cut2 destruction. The assembly of Cut9 into the 20S complex requires functional Nuc2, and vice versa. The size of fission yeast APC/cyclosome is similar to that of higher euka...

Correction: LATS1 and LATS2 Phosphorylate CDC26 to Modulate Assembly of the Tetratricopeptide Repeat Subcomplex of APC/C

In budding yeast, the Mitotic Exit Network (MEN) regulates anaphase promoting complex/cyclosome (APC/C) via the Dbf2-Cdc14 signaling cascade. Dbf2 kinase phosphorylates and activates Cdc14 phosphatase, which removes the inhibitory phosphorylation of the APC/C cofactor Cdh1. Although each component of the MEN was highly conserved during evolution, there is presently no evidence supporting direct...

The budding yeast U5 snRNP Prp8 is a highly conserved protein which links RNA splicing with cell cycle progression.

The dbf3 mutation was originally obtained in a screen for DNA synthesis mutants with a cell cycle phenotype in the budding yeast Saccharomyces cerevisiae. We have now isolated the DBF3 gene and found it to be an essential gene with an ORF of 7239 nucleotides, potentially encoding a large protein of 268 kDa. We also obtained an allele-specific high copy number suppressor of the dbf3-1 allele, en...

Thalassemic Mutations in Southern Iran

Background: Approximately 180 mutations have been described in β-thalassemia worldwide with specific spectrum in each ethnic population. This study determines the spectrum and the frequency of β-thalassemia mutations in patients with β-thalassemia trait and sickle cell-β-thalassemia. Methods: Fifteen compound heterozygous sickle cell thalassemia (SCT) and 23 β-thalassemia trait patients were st...

تعیین جهش در اگزون 8 ژن p53 در بیماران مبتلا به تومور مغزی از نوع آستروسایتوما

Background: Most studies have shown that there are association between the development and malignancy of brain tumors and tumor suppressor genes and oncogenes. The aim of this project was to investigate the P53 gene mutations in exon 8 in patients with astrocytoma type’s brain tumor. Methods: In this present survey, The DNA isolation from 30 samples of brain tissue was done by phenol-c...
